1. The Collection software suddenly crash and quit in the middle
of my run and the sequencer is still running, what should I do?
Please remains calm and don't shut down the sequencer because that will result in data lost. You can just reboot the Macintosh, start the collection software again and wait for a while. The collection should resume without losing data.2. The Collection software is running and the Sequencer is running but there is no image collected
This is usually caused by entering commands too fast. For example you have just finish the sample loading and press the stop button and start the run immediately before the machine fully stop. The laser sometimes stops to response because of this. You can try to listen if the laser is working (if you can!). The solution is stop the collection, wait till the machine fully stop before you start the machine again.3. Which run module is superior? 2400 or 1200?
Basically they should be almost the same. Theoretically 2400 should be better because the voltage is more than double comparing to the voltage for 1200. Since the running time is half for 2400, therefore using 2400 can save the laser life4. What can I do to speed up my sequencing reaction?
Instead of doing your sequencing tube by tube, you should consider doing batch sequencing on 96-well plate. Please refer to the "Performing sequencing reactions on 96-well plate" under "Experimental Protocols"5. If I can't see the "Run Under Analysis" folder and "Share Folder", what should I do?
Instead of manually and visually comparing the sequences, you should consider using Factura and Sequence navigator to help your analysis. Please refer to the "Analysis of sequencing data with Sequencing Analysis, Factura and Sequence Navigator" under "Analysis Protocols" for further detail.
