Performing sequencing reactions on 96-well plate

• The reaction itself is the same as normal sequencing reactions, except you do it on 96-well plate.

• Try to utilize the multi-channel pipetteman to dispense the solutions

• To precipitate the reaction in 96 well plate:
• •  Put the plate in -80 degrees for 15 minutes
• •  Centrifuge at 3000 rpm for 1 hour (Rotor: Beckman S2096)
• •  Remove supernatant (by turning the plate upside down)
• •  Wash the pellet with 70% alcohol
• •  Centrifuge again (3000 rpm for 1 hour)
• •  Remove supernatant (by turning the plate upside down)
• •  Dry the pellet in thermal cycler for 5 minutes at 94 degrees
• Add 1ul deionized-formamide dye to each well to resuspend the pellet
